Effect of high hydrostatic pressure on the barrier properties of polyamide-6 films BJMBR
Schmerder,A.; Richter,T.; Langowski,H.-C.; Ludwig,H..
Little is known about the barrier properties of polymer films during high pressure processing of prepackaged foods. In order to learn more about this, we examined the influence of high hydrostatic pressure on the permeation of raspberry ketone (dissolved in ethanol/water) through polyamide-6 films at temperatures between 20 and 60ºC. Permeation was lowered by increasing pressure at all temperatures. At 23°C, the increasing pressure sequence 0.1, 50, 100, 150, and 200 MPa correlated with the decreasing permeation coefficients P/(10(9) cm² s-1) of 6.2, 3.8, 3.0, 2.2, and 1.6. Analysis of the permeation kinetics indicated that this effect was due to a reduced diffusion coefficient. Pressure and temperature acted antagonistically to each other. The decrease in...

Effect of Hydrostatic Pressure on the Fluorescence of Tryptophan in the Presence of Metal Ions Running title: Effect of pressure on the tryptophan BABT
Bao,Chengman; Tang,Xinhui; Ye,Shuming.
ABSTRACT The effect of pressure on the fluorescence of tryptophan in the presence of metal ions was studied by fluorescence spectrometry. It was found that at 60 MPa, the fluorescence intensity of M/Trp mixtures (M represented metal ions) increased compared to that at atmosphere pressure. The relative fluorescence efficiency of M/Trp mixtures increased with pressure. When the M/Trp ratio was above 10:1, the relative fluorescence efficiency in decreasing order was Cu2+/Trp mixtures, Ni2+/Trp mixtures and Mg2+ (K+)/Trp mixtures. When the ratio was below 10:1, the decreasing order was Cu2+/Trp mixtures and Ni2+ (Mg2+, K+)/Trp mixtures. The relative fluorescence efficiency increased with the concentration of Cu2+ and Ni2+. The variation was relate to the...

Etude de la diversité microbienne des bassins hypersalés anoxiques profonds de la Mer Méditerranée orientale ArchiMer
L'Haridon, Stephane.
Since 1983, the existence of Deep Hypersaline anoxic basins (DHABs) has been revealed in the Eastern Mediterranean basin. These basins which represent extremes environments (anoxic, hypersalty, dark, high hydrostatic pressure)  have  been  under  investigation  and  increasing  interest  the  last  fifteen  years.  The  molecular  approaches revealed  the  presence  of  active  and  uncultivated  microbial  communities  in  the  DHABs  with  two  main  metabolic processes  such  as  methanogenesis  and  sulphate-reduction.  Until  now,  no  representative  strain  of  the  different uncultivated lineages thriving in the DHABs (ex : lineages MSBL, Mediteranean Sea Brine Lake) or involved in the two processes  have  been  yet  cultivated.  The  main ...

Isolation and physiological characterization of two novel, piezophilic, thermophilic chemolithoautotrophs from a deep-sea hydrothermal vent chimney ArchiMer
Takai, Ken; Miyazaki, Masayuki; Hirayama, Hisako; Nakagawa, Satoshi; Querellou, Joel; Godfroy, Anne.
P>Two novel, thermophilic piezophiles, capable of chemolithoautotrophic growth, are successfully cultivated and isolated from a black smoker chimney at the TAG field (Mid Atlantic Ridge: MAR) by using a piezophilic cultivation technique. Both strains (strains 106 and 108) represent dominant cultivated populations of the microbial communities in the chimney surface habitat. Strain 106 represents typically thin, long spiral cells under the piezophilic growth condition but short bent cells under the non-piezophilic condition. It is a strictly chemolithoautotrophic gammaproteobacterium using reduced sulfur compounds as the electron donors, and nitrate and O-2 as the electron acceptors. Based on the 16S rRNA gene sequence, strain 106 would represent a novel...

Long-term maintenance and public exhibition of deep-sea Hydrothermal fauna: the AbyssBox project ArchiMer
Shillito, Bruce; Ravaux, Juliette; Sarrazin, Jozee; Zbinden, Magali; Sarradin, Pierre-marie; Barthelemy, Dominique.
The AbyssBox project aims to provide the first permanent public exhibition of live deep-sea hydrothermal fauna maintained at in situ pressure. AbyssBox is a pressurized aquarium designed to function permanently. Here we present details of the project after the public exhibition functioned for more than three years at Océanopolis aquarium in Brest, France. We also describe the AbyssBox pressure aquarium, and provide data and observations on vent shrimp (Mirocaris fortunata) and crabs (Segonzacia mesatlantica) that were sampled from 1700 m depth at the Lucky Strike vent field (Mid-Atlantic Ridge) during different cruises. While mortalities exceeded 50% during the first days following sampling, the remaining animals appeared to acclimate fairly well. Some...

Measuring the biological impact of drilling waste on the deep seafloor: an experimental challenge ArchiMer
Lelchat, F.; Dussauze, M.; Lemaire, P; Theron, M.; Toffin, Laurent; Le Floch, Stephane.
The depletion of traditional oil fields is driving the oil & gas industry to explore new exploitation sites previously considered as unprofitable. Deep-sea oil fields represent one of these new areas of exploitation. Well drilling during exploration and production operations generate large quantities of drilling waste whose biological impact on the deep-sea floor remains largely unknown. Because of the harsh abiotic factors characterizing this environment, the evaluation of this impact remains challenging. High hydrostatic pressure is the prominent factor which will affect in-situ biological processes. This review will examine the feedback on the various strategies used to evaluate the biological impact of deep-sea drilling waste deposition as well as...

The biological effects of high-pressure gas on the yeast transcriptome BJMBR
Matsuoka,H.; Suzuki,Y.; Iwahashi,H.; Arao,T.; Suzuki,Y.; Tamura,K..
The aim of the present study was to examine the feasibility of DNA microarray technology in an attempt to construct an evaluation system for determining gas toxicity using high-pressure conditions, as it is well known that pressure increases the concentration of a gas. As a first step, we used yeast (Saccharomyces cerevisiae) as the indicator organism and analyzed the mRNA expression profiles after exposure of yeast cells to nitrogen gas. Nitrogen gas was selected as a negative control since this gas has low toxicity. Yeast DNA microarray analysis revealed induction of genes whose products were localized to the membranes, and of genes that are involved in or contribute to energy production. Furthermore, we found that nitrogen gas significantly affected the...

Thermoplastic composite cylinders for underwater applications ArchiMer
Davies, Peter; Riou, Luc; Mazeas, Florence; Warnier, Philippe.
A study of thermoplastic matrix composites has been performed to investigate their use in underwater applications such as oceanography, submarine, and sub-sea offshore structures. This article first presents six candidate materials. Results from simple mechanical and seawater aging screening tests on flat specimens are then described. Glass/epoxy and carbon/epoxy composites are used as reference materials. Two materials emerged from this process, glass/PEI and carbon/PEEK. Cylinders of both were manufactured and subjected to hydrostatic pressure tests, and results are compared to those for glass/epoxy and carbon/epoxy cylinders of similar geometry. The carbon/PEEK material appeared most promising. It resisted pressures in excess of 90 MPa and was retained...
